We know that good sleep is optimal for health, yet the cause of sleep disturbance is still a mystery to most of us. Understanding genetic risk factors for sleep disturbance can help create a sleep schedule and routine that works for you.

For example, those who consider themselves “morning people” may have inherited that trait. The ‘A’ allele of SNP rs73598374 in the ADA gene is associated with a 35% reduction in ADA activity which leads to an accumulation of adenosine, as it is not cleared while sleeping. While the ‘A’ allele can improve the quality of deep sleep as excess adenosine remains present, a feeling of sleepiness, lethargy, or fatigue can persist into the morning due to residual adenosine activity. In these people, enhanced deep sleep is the trade-off for morning grogginess.

In addition, The risk ‘C’ allele of rs687577 in the GRIA3 gene has been associated with depression and reduced sleep duration, with an especially strong association seen in women. No mechanism is described, but we can hypothesize that in those with the ‘C’ allele GRIA3 activity is increased, either due to an increase in the numbers of receptors present or an increase in the excitatory effect which is induced by GRIA3 upon glutamate binding.
